品系来源

NOG小鼠由日本实验动物研究所(CIEA)的Mamoru Ito培育而成。Ito博士领导的小组在CIEA通过杂交NOD/scid小鼠和γ-链IL-2受体(一些细胞因子的受体)敲除(IL2rγKO)小鼠成功建立了一个免疫功能严重不全的小鼠品系,称之为NOG(NOD/Shi-scid IL-2R小鼠品系)小鼠。与NOD/scid小鼠相比,NOG小鼠的人体细胞和组织移植存活率显著提高,同时能够植入更高比例的正常或癌变人类细胞和组织。

特性:

  • NOD背景,携带SCID突变,IL-2受体γ链基因突变
  • NOD背景导致小鼠NK细胞功能低下,补体和巨噬细胞活性降低,且NOD背景小鼠巨噬细胞表面的信号调节蛋白sirpα,对人CD47具有高亲和力,利于人类细胞/组织的移植
  • SCID突变系导致T和B淋巴细胞功能缺失
  • IL-2受体γ链基因突变引起NK细胞和DCs功能缺陷(即IFN-γ)
  • 具备无渗透率的优势 寿命长,可达1.5年
  • 自发性淋巴瘤发生率非常低(Kato C, et al., 2009)
  • 对辐射较敏感,人源造血干细胞CD34+移植,X射线的辐射剂量不要超过2.0Gy
